Stheno is one of the two Gold Assassins available at the game’s start. She has a decent ATK stat, although Assassins’ low damage multiplier hurts her quite a bit in this regard. She also possesses high innate NP gain and critical star generation rate, though the latter isn’t really visible due her Quick card’s low hitcount. Both of Stheno’s skills are crowd control type, with her first skill decreasing an opponent’s NP gauge and her second skill applying charm to a male Servant. Another thing of note is Sthenos’ high debuff resistance rate. At 50% base, it makes her much harder to debuff compared to other Servants.
Her Noble Phantasm is probably Stheno’s greatest weakness. Dealing no damage, instead it has a chance to instantly kill a male opponent. It also has a 100% chance Charm, and applies a defense buff to the target. The fact that everything save the defense buff are exclusively anti-Male is already significant disadvantage, but this is made worse by instant kill’s reliance to the opponent’s hidden death rate stat. Although her NP data says that it has a 100% chance to instantly kill, this is actually more on the lines of 100% times the opponent’s deathrate - which is usually somewhere around 20-30% for Servants, and even less on bosses.
After strengthening quest (currently unavailable on the EN ver.), she gains Whim of the Goddess A, a skill which increases the party’s attack, with additional bonus for Divine Servants. This makes her a decent Divine Servant support, although it doesn’t really solve her main problem, which is her Noble Phantasm.
Pros
High NP gain.
Crowd Control functions.
Divine support skill (Unavailable on the EN ver).
Cons
Noble Phantasm’s instant kill is unreliable.
Chance-based NP drain.
Strategies & Tips
Stheno may be used when there is no better Assassin, as her normal attacks still deal decent damage against Riders. Outside of this, she is only used in Male-only stages, especially in teams revolving around having the opponent constantly under Charm/Stun effects.
Noble Phantasm
Smile of the Stheno is a non-damaging Noble Phantasm, which has a chance to instantly kill a male opponent. It also applies a Charm if the target is male alongside a defense debuff. Stheno is able to perform this Noble Phantasm rather often due to her high NP gain rate; however, the Noble Phantasm itself is rather unreliable due to its nature. However, combined with her Charm skill, it does allow Stheno to charm male opponents at a much faster rate than any other Servant.
A natural critical damage dealer, Gilgamesh is able to make use of the critical stars Stheno provides. Additionally, her crowd control functions are also beneficial due to his lack of defensive skill.
After the servant strengthening quest (currently unavailable on the EN ver), Being a Divine Servant, Stheno’s third skill increases his damage greatly.
A fellow Gorgon Sister, Euryale also possesses anti-Male properties, and is a Divine Servant. As such, a Stheno - Euryale combination is able to severely hinder male opponents. With another Euryale/Stheno and leveled skills, it is even possible to apply a permanent Charm lock on a male target.
After the servant strengthening quest (currently unavailable on the EN ver), being a Divine Servant and possessing a critical absorb skill as well as a critical damage buff, Proto Cu benefits from both Stheno’s third skill and her high critical star generation rate.

Upgrade Priority
Vampirism C: An NP drain skill, which fills Stheno’s own NP gauge. It is recommended to alternate between leveling this skill and Siren Song to increase their hit rate. As for which skill should be prioritized, it depends on the way you wish to use Stheno. Vampirism is better for general usage, but Siren Song is better for male-only scenarios.
Siren Song A: This skill has a chance to apply Charm on a male target. If Stheno is to be used in teams focusing on alternating Charms, this skill should be leveled first for the cooldown reduction.
[JP-Only] Whim of the Goddess A: This skill should be raised before her other skills, as it is extremely flexible and beneficial for most of Stheno’s possible allies - even non-Divine ones. However, it should be noted that Siren Song benefits more from the cooldown reduction.

Stheno’s QQQAB deck and high critical star generation rate allows her to generate a good amount of stars. The lack of Arts cards is also not a problem due to her high NP gain rate. However, only having one Buster card makes it impossible for her to create NP Buster Brave chains, and thus limits her to more supportive roles.
[JP-Only] As not all Divine Servants are Quick-oriented, her lack of Arts cards can be somewhat of a hindrance.
